ABSTRACT

The objective of the present investigation is to develop simple paper and micro thin layer chromatographic enzymatic methods for detection and seperation of Zinc sulphate in fresh water. Though there are many non-enzymatic instrumental methods for heavy metal analysis, the procedures are costly and time taking. In the enzymatic methods of detection, as the reaction is specific, all the interfering substances can be avoided. Simple concentration by evaporation is followed for improving the sensitivity of the samples. The zinc sulphate can be detected by succinate dehydrogenase inhibition using 2-(4-iodophenyl 1-3-( nitrophenyl)-5 phenyltetrazolium chloride (INT)- sodium succinate- N- methylpheazonium methosulphate (PMS) mixture as the chromigenic reagent. Microgram amount of zinc sulphate can be use seperated and indentified by using this methods.
