FROM RAUVOLFIA TETRAPHYLLA

ABSTRACT

ß-Glucosidase (EC 3.2.1.21) was extracted from different parts of Rauvolfia tetraphylla and was purified using ammonium sulphate fractional precipitation, sephadex G-25 chromatography. From studies, it was observed that mature leaf 1 has the highest activity of crude enzyme in Rauvolfia tetraphylla. The purification of enzyme indicates that the average value of protein was highest at the step of ammonium sulphate precipitation (16.79 mg).The molecular weight of different bands in Rauvolfia tetraphylla was found in the range of 19.4KDa to 92.257KDa. The optimum activity of ß-glucosidase was found to be 40ºC and stability was found at 20-30ºC at room temperature. The activation with FeSO4 is due to Fe++ ions because FeCl3 do not activate the enzyme.
