ALGINATE FOR L-ASPARAGINASE PRODUCTION

ABSTRACT

A mutant strain Streptomyces gulbargensis mu24, immobilized in calcium alginate was explored for the production of L-asparaginase using groundnut cake extract as substrate/medium. The medium with initial pH 8.5, inoculated with free and immobilized cells was subjected to fermentation at 40oC at 200 rev/min. In an immobilized cell system, the enzyme production was enhanced by 18.5% (53 IU/mL) compared to the conventional free-cell fermentation (44.7 IU/mL). The immobilized cells were subjected to repeated batch fermentation process to determine their reusability. The entrapped cells retained their ability to produce L-asparaginase over five cycles and the activity remained between 32 and 53 IU/mL throughout the cycles. The calcium alginate immobilized cells of Streptomyces gulbargensis mu24 can be proposed as an effective biocatalyst for repeated usage for maximum production of L-asparaginase.
