ABSTRACT

Advances in molecular biology techniques have provided the basis for uncovering virtually unlimited number of DNA markers. The discovery of PCR to amplify a set of randomly distributed loci in any genome - facilitated the development of variety of genetic markers. The technique is relatively fast, cheap and easy to perform and   has been chosen in the present study to assess the genetic variation between male and female Zebrafish, Danio rerio albino. Morphometric analysis showed significant variations in parameters such as head length, total body length, fork length, body depth and pre-fin length between both the sexes. Further among seven different 10mer primers tested, three primers viz., 5’ GTGACGTACG 3’ (OPA 08), 5’ GTGATCGCGG 3’ (OPA 10) and 5’ ACGGCCGACC 3’ (OPE 03) showed amplification of DNA in both males and females showing sex-specific band patterns, while 5’ GAGGATCCCT 3’ (OPF 02) and 5’ CCTGATCACC 3’ (OPF 03) showed DNA amplification only in females exhibiting six and seven unique bands respectively and thus the specific molecular markers. These sex-specific molecular markers thus identified for the first time in an albino strain of zebrafish, an universal model will have high application potential in progeny testing during genetic manipulation studies.
